摘要:锰(Mn)是地球上储量最丰富的金属之一。它在正常细胞活动中不可或缺,但锰的过量暴露会引起毒性效应。神经元较之其他细胞是Mn诱导的毒性中更具易感性的。Mn在脑组织中的累积会导致伴随有帕金森样症状的的锰中毒。过去十年中发现了大量锰转运蛋白,这提高了我们对锰在细胞内外的转运的认识。然而,锰诱导的神经毒性的机制仅揭示了一部分,仍需进一步全面揭示锰诱导的毒性效应。本篇综述涵盖了锰引起的从秀丽隐杆线虫到人类的神经毒性,并探究了未来可能有助于揭示其神经毒性机制的发展方向。

3. Reproductive and paternal mediated developmental toxicity of benzo(a)pyrene in adult male Wistar rats

摘要:镉(Cd)作为一种人类致癌物可能与表观遗传学改变有关。然而,其在表观遗传学中的潜在机制和作用仍不明确。DNA甲基化是调控细胞表达的表观遗传学机制之一。我们前期的体内试验表明用20nmolCd处理4周,大鼠肝组织caspase-8基因启动子区表现出高甲基化。本次研究将揭示DNA甲基化是否也参与到Cd诱导的FL83细胞增殖过程中。当FL83细胞仅暴露于低剂量Cd(0.085 μM) 14天时,细胞增殖和DNMT甲基转移酶表达和活性增加,而癌基因caspase-8蛋白的mRNA和蛋白显著减少,并伴有细胞凋亡的显著增加和细胞侵袭转移的增加。此外,Cd暴露的FL83细胞的caspase-8基因启动子区出现高甲基化,这与我们的体内试验结果相符。一种DNA甲基化抑制剂5-氮杂-2'-脱氧胞苷(5-aza-dC)抑制了caspase-8参与的Cd诱导的细胞增殖、侵袭和转移过程。以上结果表明低剂量Cd可能诱导caspase-8基因启动子区高甲基化,导致其表达下降,从而促进细胞增殖、侵袭和转移以及减少凋亡。这些过程均在Cd介导的致癌过程中发挥作用。

摘要:我们先前的研究揭示了交通相关PM2.5暴露对秀丽隐杆线虫,一种非哺乳类毒理动物模型的损伤效应。本次研究中我们通过探究遗传方式、基因表达模式、肠道自体荧光、氧化应激和肠粘膜通透性来研究交通相关PM2.5暴露经由体内胰岛素通路对线虫的分子调控的毒性机制。在基因编码的胰岛素信号通路中,PM2.5的持续暴露引起daf-16和daf-18基因表达水平降低以及daf-2, akt-1,和pdk-1基因表达水平的升高。与此同时PM2.5暴露提高DAF-16::GFP的核定位。编码胰岛素受体的daf-2基因变异使PM2.5暴露的线虫的肠道发育和功能出现缺陷。与之相反的是,编码FOXO转录因子的daf-16基因变异对PM2.5暴露的线虫的肠道发育和功能造成更为严重的损伤。此外DAF-16可能通过诸如SOD-3,一种锰-超氧化物歧化酶等下游目标来调节PM2.5的肠道毒性。因此,胰岛素信号通路可作为PM2.5交通暴露对线虫毒性的一种重要的分子基础。
