1. Computational toxicology, friend or foe?

摘要:减少动物试验并应对环境、饮食或服药治疗疾病的化学品暴露对健康威胁的评价已成为重要公共卫生问题。计算机模拟方法因能快速高效地处理数以千计的化学结构而引人瞩目。因此,在过去十多年里受到许多人的关注并投身于此,尤以制药工业评价中应用最为显著。在制药工业中新药开发价格高昂并且由安全问题所导致的失败率较高。许多已发布的应用方法已经广泛覆盖了不同领域的人类健康和环境健康问题。正如所有新技术一样,这些新方法有被广泛应用的趋势,但可靠性和性能也可能被夸大。那么这些计算机模拟方法到底有多好呢?本文旨在为计算毒理学进展做一个概述,来阐明这些方法中存在的一些局限性以使得这些有价值的工具能够被正确地应用和理解。

13. Development and use of in vitro alternatives to animal testing by the pharmaceutical industry 1980–2013

摘要: 我们调查了1980到2013年间制药工业在临床前安全性试验中使用体外技术(包括计算机模拟)的情况。通过调查英国制药工业协会(ABPI)成员企业以及从非临床生物研发专家网络(NaBDEN)获取1980到2005年间每隔5年和2008年后每年用体外和模拟试验筛查过的化合物的信息。每个实验都要求用1分(低)到5分(高)评价其效用值。4家制药企业和3家合约研究机构(CROs)对本调查作出回应,提供了超过895 000并覆盖全部年份和方法的数据。总体来说,在1980到2013年间,制药企业中利用体外实验的情况呈平稳上升趋势;事实上在去年的调查期间(2013)所有报道显示大于20%的体外试验已经实施;超过70%的体外实验在2010年后实施。体外试验在2012年的使用达到了每年超过19,000的峰值;超过99%的使用集中在ADME、安全药理学以及遗传毒性这三个主要领域,上升趋势变化在ADME、安全药理学及遗传毒性三个主要方面尤其显著,并可由国际协调会议指南相关部分的采用时机进行解释。也可通过由低(眼刺激性-荧光素漏出)到高(遗传毒性-Ames实验和皮肤刺激性-人重组皮肤模型EpiSkin/Epiderm))的效用认知评分来解释这种趋势。总之,本研究的数据展示了药物研发领域中动物实验体外替代法发展运用的巨大进展与持续增长趋势,这为提高药物研发成功率以及恪守3Rs原则提供了新的契机。
