有毒金属污染物铅(Pb)的职业暴露和环境暴露是全球普遍关注的问题。本研究旨在探究锂(Li)对铅诱导的体内外损伤的保护作用。我们用锂(250 mg/kg体重,i.p.)处理3月大的小鼠并在2小时之后给予含铅(20 mg/kg体重,i.p.)的水共染毒两周。铅暴露诱导小鼠出现显著的多器官形态损伤,如肝脏、肾脏和脾的肿胀坏死。免疫组化结果表明铅暴露小鼠海马内新生细胞和未成熟神经元的数目明显低于用生理盐水或Li处理的对照组。此外,铅暴露小鼠的新生细胞分化成角质细胞的比例升高,而分化为神经元的比例降低。与用生理盐水或Li处理的小鼠,铅暴露小鼠的新生细胞存活率更低,认知能力也降低。有趣的是,提前施以Li可降低Pb诱导的病理和神经损伤。Li处理也可预防 Pb暴露所致的海马神经元生成减少。另外,用Li预处理可明显提高由Pb诱导的海马p-GSK-3β (Ser9)和microRNA-34c水平的下降。综合以上结果我们揭示Li拮抗Pb暴露所致的损伤效应。

氯氰菊酯农药已被证实可诱导成年哺乳动物的神经毒性。探究氯氰菊酯在大脑发育过程中的毒性及其在神经元中的作用机制仍然重要。我们在近期研究中利用神经生长因子(NGF)分化PC12细胞识别出主要上调的miRNAs是miR-200家族,该家族miRNA能调节PC12细胞分化为神经元。在本研究中,我们比较了氯氰菊酯在未分化的PC12细胞和神经元样分化PC12细胞间的毒性差异,并研究miR-200家族在氯氰菊酯诱导的神经元细胞死亡过程中的作用。我们的结果表明非细胞毒性浓度的氯氰菊酯选择性诱导miR-200家族和分化PC12细胞的凋亡,而在未分化P12细胞中为未能观察到明显改变。此外,本研究揭示氯氰菊酯通过增加分化PC12细胞中的P53水平进而诱导miR-200生成。PC12细胞中miR-200表达和P53水平呈相关关系,进一步研究发现BCL2是miR-200b/c的靶向蛋白,BCL2蛋白的下调可影响氯氰菊酯诱导分化PC12细胞的凋亡。利用miR-200家族抑制剂进行的逆转试验进一步证实miR-200家族对于氯氰菊酯暴露的分化P12细胞的凋亡效应的影响。综上所述,本研究表明相对于未分化的PC12细胞,分化了的PC12细胞对氯氰菊酯更加敏感;由P53诱导的miR-200家族可调控氯氰菊酯介导的神经元样分化PC12细胞的凋亡。
